Targeted methylation analysis

Targeted methylation analysis will be applied during the tuning and validation phases of the MAMELI project to focus on specific REs identified in the discovery phase.

A custom-designed targeted panel will be created to detect methylation at single genomic positions of the previously identified REs with high specificity. This approach will enrich for those REs found to be modifiable during the discovery phase, ensuring precise quantification of their methylation status. Methylation sequencing will be performed using the Next-Seq2000 Illumina platform, which generates detailed methylation data for each targeted RE.

The methylation data obtained from this targeted sequencing will be critical for the development and validation of the MAMELI algorithm, which aims to model the relationship between RE methylation and exposome factors.
